Abstract

Cancer is the rapid creation of abnormal cells that grow beyond their usual boundaries, and which can then invade adjoining parts of the body and spread to other organs. Worldwide, an estimated 19.3 million new cancer cases (18.1 million excluding nonmelanoma skin cancer) and almost 10.0 million cancer deaths (9.9 million excluding nonmelanoma skin cancer) occurred in 2020. Cancers have the ability to develop resistance to traditional therapies, and the increasing prevalence of these drug resistant cancers necessitates further research and treatment development. This research paper deals with formulation of thiolated konjac gum chitosan nanoparticles (CS-NPs) for the mucoadhesive drug delivery of the colon cancer, which helps to conquer acquired drug resistance. In this work three optimization variable concentrations like particle size, zeta potential and PDI were selected using full factorial design using two levels. The results showed that we have statistical optimization formulation which have least particle size, drug release characteristics verified using formulation.

 

Background:

Acquired drug resistance is becoming common during cancer chemotherapy and leads to treatment failure in the clinic. To conquer acquired drug resistance, nanotechnology has been employed to deliver the drug. In this dissertation, we formulate the thiolated konjac gum chitosan nanoparticles (CS-NPs) for the mucoadhesive drug delivery of colon cancer.

 

Methodology:

Many studies have been conducted in recent years to develop bioadhesive/mucoadhesive managed release systems. In this work, the thiolation of the Konjac gum was done with an esterification reaction, the konajac gum was esterified with thioglycolic acid (TGA 80 percent) in the presence of hydrochloric acid and precipitated with the help of Ethanol. The chitosan nanoparticles are prepared by the ionic cross-linking method. The CNPs is prepared by Chitosan are cross-linked with sodium tripolyphosphate by the dropwise addition of TPP in the CS-TK-GF solution with the syringe and wash with the water.
