Abstract

Mastitis is a mammary gland disease that is commonly caused by bacterial agents. Mastitis is very common disease of bovine animals in both developing and developed countries. As compared to develop countries economic losses caused by mastitis are higher in Pakistan because prevention strategies of mastitis are not so much established in Pakistan. Mastitis caused by Genus Staphylococcus is familiar and economically biggest worry where dairy farming is performed. An infected udder is main reservoir of bacterium Staphylococcus. Staphylococcus is opportunistic and contagious bacteria that causes sub clinical and clinical mastitis in dairy animals. This analysis was performed to estimate incidence of mastitis and  to isolate and identify genus Staphylococcus in dairy cows and buffaloes in particular dairy farms of District Abbottabad. A total of 200 milk samples of dairy cows and buffalo specie taken in sterile condition were investigated for presence of mastitis by using a California mastitis test (CMT). Genus Staphylococcus was isolated and identified by culturing CMT positive bovine milk samples on Nutrient agar which was followed by Gram staining and Biochemical test. In present study overall prevalence of mastitis found to be 54% (108/200). According to CMT out of the 200 samples examine for mastitis 92 samples (46%) were negative, 26 (13%) were trace-positive, 28 (14%) were weak-positive, 28 (14%) were distinct-positive and the rest 26 (13%) samples were strong positive. Overall prevalence of mastitis at cow level found to be 68.75% (44/64) and prevalence at buffalo specie level found to be 47.05% (64/136) by using California mastitis test. Among these 108 CMT positive samples 106 samples were found to be culture positive and bacterial organisms were isolated from these culture positive samples. From these 106 culture positive samples a total of 72 (67.92%) samples were positive for genus Staphylococcus. In Mastitis positive milk samples genus Staphylococcus was found in higher percentage.
